Metals and ceramics have found widespread application in orthopaedics and dentistry as biomaterials such as artificial joints, osteosynthesis devices, intervertebral cages or dental implants. The design of their material surfaces plays a key role to provide rapid integration of the specific implant into the surrounding bone tissue and to ensure their long-term functionality. In addition to a proper osseointegration of implants their protection against bacterial infection becomes more and more important due to the problem of implant-related microbial infections. There exist various concepts to tailor biomaterial surfaces with regard to their application requirement.
In this context we report on the development of new coating systems based on biodegradable and highly biocompatible functional materials acting as carriers for both osteogenic agents like cell adhesion peptides, growth factors, and low-molecular weight drugs as well as antimicrobial agents. Novel polysaccharide-based coating systems allow the selective immobilisation of cell adhesion stimulating peptide sequences onto metallic or ceramic implants. A further approach focuses on the manufacture of sparingly soluble salts of low and high molecular weight bioactive agents providing a controlled and sustained release of the bioactive agent over time. Results on in vitro and in vivo-testing are presented and the clinical application potential of the new coatings is discussed.
